GENOTYPE DIVERSITY OF THREE GENERATIONS OF KURUMOI RAONBOW FISH (Melanotaenia parva) BY AQUACULTURE

Kurumoi rainbowfish (Melanotaenia parva) is endemic fish from Kurumoi Lake, Papua, Indonesia. Research and Development for Ornamental Fish Culture, Depok has successfully domesticated and produce Kurumoi rainbowfish for several generations. This fish is breeding naturally, so inbreeding probability is high. It is important to know characteristic of genetic resources Kurumoi rainbowfish that had been inbreeding. The aim of this research is knowing genotype diversity of three generations of Kurumoi rainbowfish for genetic quality. Characterization of RAPD (Randomly Amplified Polymorphic DNA) method used to determine quality genetic resources this fish. Fin organ used as a sample, with sample of 10 individuals/generation of total from three generations. Amplification used three primers OPA18, OPZ5, and OPZ13. The result from the study is the lowest genetic diversity found in first generation and the highest found in third generation, with polymorphism range at 69.2% -76.9% and heterozygosity range at 0.2091 - 0.2476 The longest genetic distance is between the first and third generation (0.1855) (Table 1). Similarity between three generatiaons of kurumoi rainbowfish show in the dendogram (Figure 1).  Genotype diversity from three generation had enhancement but in statistic is not significantly different between three generations kurumoi rainbowfish (Table 1). In order to improve the genotype diversity, made efforts to improvement of the genetic quality through selective breeding and crossbreeding.
