A LOW-COST SYSTEM FOR KINETIC ANALYSIS OF CALIGUS ROGERCRESSEYI; NEW FOCUS ON PHARMACOLOGICAL STUDY OF DISEASE

The study of the Caligus rogercresseyi (caligus ) is a complex challenge in our country. Caligus generate important effects on salmon production in Chile and other countries. One of the problems in the study of caligus is the cost to test a new compound. The development and generation of in vitro model of infection are expensive because of the biosecurity protocol in general and animal care. Our objective was to develop a low-cost system for evaluate the effect of different drugs in caligus . We previously used a low-cost software (ImageJ) for observation of kinetic parameters in sperm cells, for follow "particles" in microscope with video recording. We took caligus on different lifecycle stages (copepodites) and in an inverted microscope we recorded the movement from samples. Then we used a modified protocol for ImageJ analysis (CASA) and obtained the kinetic parameters. The caligus were observed under control condition (seawater at 15°C) or experimental condition (tap water dilution 0.1 at 100 V/V). Our results suggested that we can use ImageJ plugin (CASA) as a low-cost system for screening a potential compound that have the potential to alter the physiology of Caligus rogercresseyi, in particular, the kinetic of copepodites, and compare the pharmacological effect of a new drug before to do in vitro or in vivo study.
