PRODUCTION TECHNIC OF BIOFLOC HETEROTROF BACTERIAS IN LOW COST BACTERIAL CULTURE MEDIA

Lysinibacillus fusiformis strain xfm5 CB1, Bacillus sp. CB3 and  Bacillus subtilis strain Dm B4 16  CB4 are indigeneus biofloc heterotrof bacterias.  Based on research of Research and Development Institute for Fresh Water Aquaculture (BPPBAT), Bogor those biofloc bacterias optimum growth on  28-380C temperatue, normal pH range, good synergize, non pathogen, 70 - 98% reducing levels of ammonia, 7-9 days probiotic reapply on field aplication and floc volume reach until 60 ml/L.  In order to developing the potential biofloc probiotic to be commercial using low cost bacterial media culture and supporting the National probiotic, researched has been done. The purpose was to find out low cost of bacterial culture media for mass technic production. In the present experiment, the bacterial culture media were  tofu liquid waste (TLW) and molases. Probiotic was also tested for sterility, viability, biological safety level  until end product. Researched was done in laboratorium and field levels on tilapias culture ( two periods ) with biofloc system.

Results showed that bacterial culture media compositions are TLW 15% + molases 5% which high and balance of total bacterias colonies and (Table 1). The analysis of  growth bacterias population at 0 to 3 month was 109 CFU/ml,  no bacteria contaminat and non pathogen.The optimum of packaging probiotic volume  are 0,5 and 1 liter, using plastic or glass bottle and  room temperature storage. Field results showed that bacterias are non pathogen, optimum floc volume  18,5ml/L and tilapias survival rate 80% (Table 2). Water quality parameters ( temperature, pH, DO, CO2 ,NO2, NO3, NH3 and  alkalinity) were optimum  for fish and sufficientcy for growing of biofloc heterotrof bacterias      .
